Background
Caspase proteins are a family of cysteine proteases that are key mediators of programmed cell death or apoptosis (1). The precursor form of all caspase proteins is composed of a prodomain, and large and small catalytic subunits. The active forms of caspases are generated by several stimuli including ligand-receptor interactions, growth factor deprivation and inhibitors of cellular functions. All known caspases require cleavage adjacent to aspartates to liberate one large and one small subunit, which associate into tetramer to form the active enzyme. Caspase-1 (ICE, IL-1b converting enzyme) is similar to the cell death gene CED-3 of Caenorhabditilis elegans and regulates multiple proinflammatory cytokines, including interleukin-1b and interferon-gamma-inducing factor.

Antigen
A synthetic peptide corresponding to amino acids 371-390 RKVRFSFEQPDGRAQMPTTE of human caspase-1 was used as immunogen. This antibody will cross-react with mouse and rat Caspase-1, based on NCBI BLAST.

Application Notes
The antibody will recognize full-length Caspase-1 and cleaved caspase-1 forms that retain amino acids 371-390 of the Caspase-1 protein.

Western blot analysis of Caspase-1 using a Caspase-1 monoclonal antibody. Human HeLa (A) and mouse NIH3T3 lysate probed with Caspase-1 antibody at 0.5 ug/ml and 2 ug/ml, respectively.

Caspase-1 Antibody IMG-5028

Immunohistochemical analysis of Caspase-1 using Caspase-1 monoclonal antibody. Formalin-fixed, paraffin-embedded sections of human colon cancer stained for Caspase-1 expression (IMG-5028) using a DAB chromogen and Hematoxylin counterstain. Nine sections, from different human patients were stained. Differential staining was observed (top right panel). Two of these sections are shown at higher magnifications (arrows and A, B). Only rare staining in A is observed. In contrast, abundant staining is observed in B.
