Background
Ca2+ plays a critical role in intracellular signaling and thus regulation of calcium balance is critical for the maintenance of this signaling function. The plasma membrane Na+/Ca2+ (NaCX) exchangers mediate Na+-dependent Ca2+ efflux in a wide variety of cell types (reviewed in 1-3). NaCX can move Ca2+ either into or out of cells, depending on the net Na+, Ca2+, and K+ gradient across the membrane. In mammals, at least 5 distinct genes code for the exchangers: Three NCX (NCX1, NCX2, and NCX3), and two in the NCKX family (NCKX1 and NCKX2). NCX1 is most prominently expressed in the heart where it plays a major role in excitation-contraction coupling.

Antigen
A fusion protein from the N-terminus of the rabbit renal sodium calcium exchanger was used as immunogen. The antibody was purified by Protein G affinity chromatagraphy.

Application Notes
Specific for the ~120 kDa and ~160k Na+/Ca2+ Exchanger NaCX-1 protein in Western Blots with broad tissue and species specificities. Antibody dilutions and tissue load should be based on tissue type and expected expression level.
