Background
CD254 is a member of the tumor necrosis factor family. TRANCE (TNF-related activation-induced cytokine receptor) or RANKL {receptor activator of NF-kB (RANK) ligand} has been implicated in interactions between T cells and dendritic cells (1-2). CD254 (RANKL) binds to RANK on DCs, upregulates the expression of anti-apoptotic protein BcL-XL suggesting a role in dendritic cell survival (2). CD254 is also important in Tcell and Bcell maturation (3). As OPGL/ODF (Osteoprotegerin Ligand/Osteoclast differentiation factor), CD254 can both activate mature osteoclasts and mediate osteoclastogenesis (4,5). CD254 deficient mice show severe osteoporesis and complete absence of osteoclasts as a result of lack of osteogenesis (3). Mouse CD254 is a 316 amino acid protein.

Antigen
A synthetic peptide corresponding to amino acids 161-177 of mouse CD254 (AQPFAHLTINAASIPSG) was used as immunogen for this antibody.

Application Notes
The CD254 amino acid sequence used as immunogen for this antibody is 88% homologous in human and 94% homologous in rat.
