Background Sphingosine 1-phosphate receptor Edg8 is a multi-pass membrane protein belonging to the G-protein coupled receptor 1 family and coupled to both the G (i/0) alpha and G (12) subclass of heteromeric G-proteins. Edg8 acts as a receptor for the bioactive lysosphingolipid sphingosine 1-phosphate (S1P) as well as for lysolipid mediator lysophosphatidic acid (LPA) and gives diverse physiological effect on most types of cells and tissues. Edg8 may also play a regulatory role in the transformation of radial glial cells into astrocytes and affect proliferative activity of these cells. It is widely expressed in white matter tracts of brain, spleen, peripheral blood leukocytes, placenta, lung, aorta, and fetal spleen. Over expression of Edg8 leads to a lymphoproliferative disorder often associated with the autoimmune disease known as leukemic large granular lymphocyte (LGL). |
Antigen
A synthetic peptide corresponding to amino acids 15-31 (IVLHYNYTGKLRGARYQ) of human Edg8 was used as the immunogen, GenBank no NP_110387. |
Application Notes
The amino acid sequence used as immunogen is 100% homologous in human (isoform CRA_a and CRA_c), dog, rat, monkey and mouse, and 82% homologous in opossum and cow. |
