Background Autophagy is a process for the bulk degradation of cytosolic compartments by lysosomes/vacuoles. The formation of autophagosomes involves a dynamic rearrangement of the membrane for which two ubiquitin-like modifications (the conjugation of APG12p and the modification of a soluble form of MAP-LC3 to a membrane-bound form) are essential. APG10L is an E2-like enzyme involved in two ubiquitin-like modifications essential for autophagosome formation, ATG12 -ATG5 conjugation and modification of a soluble form of MAP-LC3 (MAP1LC3A), a homolog of yeast Apg8, to a membrane-bound form. The protein is ubiquitously expressed. Human APG10L gene is localized on chromosome 5q14.2. |
Antigen
A portion of amino acids 1-50 of human APG10L was used as the immunogen. |
Application Notes
The amino acid sequence used as immunogen is 100% homologous in human (isoform 2) and 87% homologous in mouse. |

Western blot analysis of APG10L in human brain lysate in the 1) absence and 2) presence of immunizing peptide 3) mouse brain and 4) rat brain using IMG-6174A at 2.0 ug/ml, 6.0 ug/ml and 4.0 ug/ml, respectively. IMGENEX’s goat anti-rabbit Ig HRP secondary antibody (IMGENEX, 20301) and PicoTect ECL substrate solution (IMGENEX, 10087K) were used for this test. |
