Background

Edg-1 is a heterotrimeric orphan receptor for lysosphingolipid sphingosine 1-phosphate (S1P) and belongs to the G-protein coupled receptor 1 family with seven putative transmembrane domains. Human Edg-1 is a highly inducible and abundant endothelial cell GPR that helps in the regulation of differentiation of endothelial cells. S1P-induced endothelial cell migration requires the PKB/AKT1 mediated phosphorylation of Edg-1. Activation of Edg-1 induces cell-cell adhesion and it also helps in modulating intracellular Ca2+ homeostasis. It is rapidly induced when endothelial cells are treated with the tumor promoter phorbol 12-myristate 13-acetate (PMA) and super induced in the presence of cycloheximide. Over expression of Edg-1 leads to cell-cell aggregation, enhanced expression of cadherins and formation of well-developed adherens junctions. Edg-1 is widely expressed in many tissues with low expression in vascular smooth muscle cells, fibroblasts, melanocytes, and cells of epithelioid origin.

Antigen
A portion of amino acids 340-400 of human Edg-1 was used as the immunogen.

Application Notes
The amino acid sequence used as immunogen is 100% homologous in human, 95% homologous in pig, horse, opossum, macaques, rheseus monkey, chimpanzee, cow, mouse, dog, rat, orangutan, 91% homologous in chicken, and 86% homologous in xenopus, platypus.
