Background
Beta-arrestin-2, or ARRB2, is an adaptor protein belonging to the arrestin family with an N- arrestin domain and a C- arrestin domain. ARRB2 is a regulatory protein involved in heterotrimeric G protein-coupled receptor desensitization and is known to regulate beta2-adrenergic receptor (beta2AR) function by enhancing beta2AR mediated nuclear translocation of ERK. ARRB2 binds to phosphorylated beta2ARs, thereby causing a significant impairment of their capacity to activate G(S) proteins. Along with AIP4, ARRB2 acts as an endosomal sorting molecule that mediates CXCR4 entry into a degradative pathway. It may also be involved in hormone-specific desensitization of TSH receptors.  ARRB2 is a Ca2+ binding protein of the retinal outer segments and binds to P-rhodopsin and is also involved in synaptic transmission in photoreceptor cells. It is widely expressed in most of the tissues, especially neuronal tissues and spleen.

Antigen
A peptide sequence corresponding to amino acids 23-40 (GKRDFVDHLDKVDPVDG) of human ARRB2 was used as the immunogen for the antibody, Gen Bank no. gb|ABG47460.1|.

Application Notes
The ARRB2 amino acid sequence used as immunogen for the antibody is 100% homologous in human, rhesus monkey, chimpanzee, mouse, rat, dog, horse and cow, and 94% homologous in pufferfish, zebrafish and xenopus.

ARRB2 Antibody WB

Western blot analysis of ARRB2 in HUVEC cell lysate in the 1) absence and 2) presence of immunizing peptide using ARRB2 antibody at 0.25 ug/ml.  IMGENEX’s goat anti-rabbit Ig HRP secondary antibody (IMGENEX, 20301) and PicoTect ECL substrate solution (IMGENEX, 10087K) were used for this test.

ARRB2 Antibody IHC

Formalin-fixed, paraffin-embedded human thyroid tissue stained with ARRB2 antibody at 5 ug/ml.

Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10 mM sodium citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min.
