Background
NALP7 is a 980 amino acid containing short NALP belonging to the NLRP family with a DAPIN domain, nine LRR (leucine-rich) repeats and a NACHT domain. NALP7 is usually induced by LPS and IL-1B in peripheral blood mononuclear cells. A PYRIN-containing APAF-1-like protein, NALP7 directly interacts with CASP1 and IL-1B and acts as a feedback regulator of CASP1/caspase-1-dependent interleukin-1 beta secretion. It may play a crucial role in cell proliferation. Expressed in numerous tissues including uterus and ovary, with low levels in heart and brain but not detected in skeletal muscle. Defects in NLRP7 are the cause of hydatidiform mole (HYDM), an abnormal human pregnancy with no embryo and cystic degeneration of placental villi.

Antigen
A synthetic peptide corresponding to amino acids 106-122 (GDAEEDSELAKPGEKEG) human NALP7 was used as the immunogen, GenBank No. NP_001120727.1.

Application Notes
The amino acid sequence used as immunogen is 100% homologous in human and rhesus monkey.

Western blot analysis of NALP7 in human liver lysate in the 1) absence and 2) presence of immunizing peptide using IMG-6357A at 0.25 ug/ml.  IMGENEX’s goat anti-rabbit Ig HRP secondary antibody (IMGENEX, 20301) and PicoTect ECL substrate solution (IMGENEX, 10087K) were used for this test.
