Background Furin, also known as Paired basic amino acid residue cleaving enzyme, is a 794 amino acid containing membrane associated protein circulating between the trans-Golgi network and cell surface. It belongs to the peptidase S8 family and Furin subfamily with a homo B/P domain. Known to represent the ubiquitous endoprotease activity within constitutive secretory pathways, Furin, a calcium-dependent serine endoprotease, can efficiently cleave precursor proteins at their paired basic amino acid processing sites (RX (K/R) R consensus motif). This proprotein convertase enzyme processes latent precursor proteins into their biologically active products and is important in the release of albumin and complement component C3 from their respective precursors and in the physiologic processing of proparathyroid hormone, von Willebrand factor, insulin receptor, TGF1B, hepatocyte growth factor, C propeptide of collagen type V, alpha 1, NOTCH signaling proteins, certain viral envelope glycoproteins and ED1. Ubiquitous expression of Furin is seen in most of the tissues. It may also play a role in tumor progression and maintainance of immune tolerance. |
Antigen
A portion of amino acids 740-790 of human Furin was used as the immunogen. |
Application Notes
The amino acid sequence used as immunogen is 100% homologous in human, wild boar, horse, mouse, chimpanzee, rhesus monkey, dog and sheep, 94% homologous in opossum and 77% homologous in chicken and Xenopus. |
