Background
DUBA, also known as OTU domain-containing protein 5, is a de-ubiquitinating enzyme belonging to the OTU5 family and DUB subfamily characterized by an ovarian tumor (OTU) domain. DUBA is a protease that specifically cleaves ubiquitin linkages, negating the action of ubiquitin ligases. This de-ubiquitinating activity is directed towards 'Lys-63'-linked polyubiquitin chains and acts via TRAF3 de-ubiquitination and subsequent suppression of type I interferon (IFN) production. DUBA acts as a negative regulator of the innate immune system. It is generally inhibited by N-ethyl-maleimide (NEM). Expression of this enzyme is seen in various tissues, including the liver and placenta, as well as in peripheral blood leukocytes. DUBA is induced by bacterial lipopolysaccharides (LPS) in bone marrow-derived macrophages.

Antigen
A portion of amino acids 550-600 of human DUBA was used as the immunogen.

Application Notes
The amino acid sequence used as immunogen is 100% homologous in human (isoforms CRA_b, CRA_c, CRA_d), chimpanzee, rat and mouse and 94% homologous in dog and cow.

DUBA Antibody

Western blot analysis of DUBA in human liver lysate in the 1) absence and 2) presence of immunizing peptide, 3) mouse liver lysate and 4) rat liver lysate using IMG-6350A at 4 ug/ml. IMGENEX’s goat anti-rabbit Ig HRP secondary antibody (IMGENEX, 20301) and PicoTect ECL substrate solution (IMGENEX, 10087K) were used for this test.
