Background CBX5, also known as Chromobox protein homolog 5, is a regulatory protein belonging to the Polycomb group proteins family with two chromo domains. CBX5 associates with chromosomes during mitosis, specifically with chromatin during metaphase and anaphase. A component of the centromeric and pericentromeric heterochromatin, CBX5 may interact with lamin B receptor (LBR) to contribute to the association of the heterochromatin with the inner nuclear membrane. CBX5 recognizes and binds histone H3 tails methylated at 'Lys-9', leading to epigenetic repression. This methyl-lysine binding protein localized at heterochromatin sites is also known to mediate gene silencing and is involved in the formation of functional kinetochore through interaction with MIS12 complex proteins. |
Antigen
A portion of amino acids 50-100 of human CBX5 was used as the immunogen. |
Application Notes
The amino acid sequence used as immunogen is 100% homologous in human (isoform CRA_a), monkey and dog and 93% homologous in mouse and rat. |

Western blot analysis of CBX5 in HCT-116 cell lysate in the 1) absence and 2) presence of immunizing peptide and 3) NIH 3T3 cell lysate using IMG-6453A at 3 ug/ml. IMGENEX’s goat anti-rabbit Ig HRP secondary antibody (IMGENEX, 20301) and PicoTect ECL substrate solution (IMGENEX, 10087K) were used for this test. |
