Background Podoplanin is a mucin type-1 glycoprotein of 40-43kd also know as T1A, gp38 or Aggrus. Podoplanin is expressed in many tumors and many normal cells, especially lymphatic epithelial cells and follicular DCs. It appears to serve as a ligand for CLEC-2. Podoplanin expression is positively correlated with tumors expressing greater invasive and metastatic potential. Podoplanin is directly involved in cell migration, aids metastases formation and tumor cell invasion of tissue (Schacht, 2005, Raica, 2008, Martin-Villar, 2010). It has also been determined that podoplanin expression is induced through Src activation of Cas (Shen, 2010). Recently there are indications of podoplanin presence on Th17 cells (Bettelli, 2011). Initial studies below indicate the presence of podoplanin on a small but distinct population of CD4+ cells from human PBMC. |
Antigen
A synthetic peptide corresponding to amino acids 38-51 (EGGVAMPGAEDDVV) from the platelet-aggregation-stimulating (PLAG) domain of human podoplanin was used as immunogen, Gene Bank no sp|Q86YL7.3|PDPN_HUMAN. |

|
Flow cytometric analysis using Podoplanin antibody. Human NCCIT cells were probed using 10 ul (0.15 ug) of Podoplanin antibody (red) and 0.15 ug of isotype control antibody (green). IMGENEX’s flow kit (10084K) was used for this test. |
