Background
The ion channels activated by glutamate are typically divided into two classes. Those that are sensitive to N-methyl-D-aspartate (NMDA) are designated NMDA receptors (NMDAR) while those activated by a-amino-3-hydroxy-5-methyl-4-isoxalone propionic acid (AMPA) are known as AMPA receptors (AMPAR). The AMPAR are comprised of four distinct subunits GluR1-4 and they play key roles in virtually all excitatory neurotransmission in the brain. The GluR1 subunit is widely expressed throughout the nervous system. GluR1 is also potentiated by phosphorylation at Ser845. In addition, phosphorylation of this site has been linked to synaptic plasticity as well as learning and memory.

Antigen
This antibody was developed against synthetic phosphopeptide corresponding to amino acids residues surrounding the phospho-Ser845 of rat GluR1. The antibody was purified by sequential chromatography on phospho- and non-phosphopeptide affinity columns.

Application Notes
An ~100 kDa band is observed in rat brain. Immunolabeling is blocked by the phosphopeptide used as antigen but not by the corresponding non-phosphopeptide. Antibody dilutions and tissue load should be based on tissue type and expected phosphorylation state expected expression level.

Metabotropic Glutamate Receptor (GluR1 [Ser845]) Antibody

Western blot analysis using 10 ug of rat brain lysate showing specific immunolabeling of GluR1 phosphorylated at Ser845 (Lane A). The labeling by the antibody was specifically blocked by the Ser845 phosphopeptide used as antigen (Lane B). The corresponding non-phosphopeptide did not block the immunolabeling (not shown).
