Background DNA Damage Binding protein 1 (DDB1) binds with CUL4 to form an ubiquitin E3 ligase that targets protein substrates for ubiquitin-dependent proteolysis (1, 2). Loss of DDB1 impedes the proteolysis of a grouping of CUL4 substrates at least in response to UV (3, 4) and CUL4 ubiquitinylation (5). DDB1 is highly conserved from yeast to mammals (3, 6). DDB1 specific antibody probes might be useful in identifying CUL4-DDB1 substrates in epigenetics applications and ubiquitin modification dependent protein function. |
Antigen
A portion of amino acids 385-435 from human DDB1 was used as the immunogen for this antibody. |
Application Notes
The amino acid sequence used as immunogen for this antibody is 100% homologous in human, panda, cow, dog, old world monkey, horse, african bush elephant, crab-eating macaque, rhesus monkey, mouse, chimpanzee, orangutan, rat, pig, and european pole-cat , 94% in guinea pig, chinese hamster, platypus, and zebra finch, 89% in carolina anole, zebrafish, mole rat, cichlid fish, and salmon, 83% in chicken, western clawed frog, and african clawed frog, 78% in wild turkey, and 61% homolgous in common grape vine, soya bean, and tomato. |

Western blot analysis of DDB1 using DDB1 antibody. Human liver tissue lysate in the 1) absence and 2) presence of immunizing peptide, 3) Hela and 4) Daudi lysate probed with 2 ug/ml of DDB1 antibody. IMGENEX’s goat anti-rabbit Ig HRP secondary antibody (20301) and PicoTect ECL substrate solution (10087K) were used for this test. |

Intracellular flow cytometric analysis of DDB1 in human HeLa and mouse RAW cells using DDB1 antibody at 0.2 ug/10^6 cells. Red represents cells with DDB1 antibody, green represents isotype control antibody (IMGENEX, 20304). IMGENEX's goat anti-rabbit PE secondary antibody (20303) was used. |
