Background Principal Names: B-cell CLL/Lymphoma 2 (BCl-2); Apoptosis regulator Bcl-2; Oncogene B-cell leukemia 2 Official Gene Symbol: BCL2 Gen Bank Accession Number: Gene ID: 596 (Human) Gene Map Locus: 18q21.3 (Human) Bcl-2, a founding member of Bcl-2 protein family, is a 25KDa integral membrane protein localized on mitochondria and endoplasmic reticulum. It is a human homolog of C.elegans ced-9 and consists of conserved BH1, BH2, BH3 and BH4 domains and a C-terminal hydrophobic domain. Bcl-2 interacts with various proteins including Bax, RAF1, Myc, BAG1 and NUR77 and is involved in functions including regulating apoptosis, maintaining B-cell memory, regulating calcium signaling and promoting cell growth. The protein also regulates a caspase-activation program independent of Cytochrome-C/Apaf1/caspase-9 apoptosome. Bcl-2 expression is up-regulated by IL-2 and its role as a proto-oncogene has been suggested in follicular lymphoma and tumor angiogenesis.

Application Notes
We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user. This antibody works well with B5 or Bouin's fixed tissues. Formalin-fixed tissues may give inconsistent results. Specificity: This antibody is specific to bcl-2 oncoprotein, encoded by agene involved in the 14; 18 chromosomal translocation. In lymphoid tissues, this antibody stains B-lymphocytes as well as T cells. Cellular Localization: Cell membrane.
