Principal Names: Hematopoietic Progenitor cell antigen; CD34 antigen; Official Gene Symbol: CD34 Gene ID: 947 (Human) Gene Map Locus: 1q32 (Human)CD34, an 110KDa monomeric cell surface glycoprotein, is a member of Sialomucin family. It is a Type I transmembrane protein selectively expressed on surface of human hematopoietic progenitor cells and endothelial cells. It consists of a heavily glycosylated extracellular domain, a transmembrane region and an intracellular cytoplasmic domain containing potential phosphorylation sites. Physiological functions of CD34 antigen include role in early hematopoiesis, cell-cell adhesion, and binding of L-selectin in high endothelial venules. CD34 antigen is frequently used for positive selection of hematopoietic stem/progenitor cells in research and transplantation. Pathological role of CD34+ stromal cells has been suggested in ALL and lung adenocarcinomas.

Application Notes
Monoclonal antibodies to CD34 can be classified into three main classes, class I, class II and class III, on the basis of differential sensitivity to corresponding CD34 epitopes to degradation by specific enzymes. The IMG-80073 is a class II monoclonal antibody which recognizes a CD34 epitope resistant to neuraminidase, and sensitive to glycoprotease and chymopapain.  We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user. Suitable for formalin/paraffin tissues. Specificity: This antibody reacts with a glycoprotein of MW 110 kD, present on the surface of hematopoietic cells i.e. myeloid and lymphoid progenitors. It stains the vascular endothelial cells. Cellular Localization: Cell membrane.
