Background The breast cancer tumor-suppressor gene, BRCA1, encodes a protein with a BRCT domain, a motif that is found in many proteins that are implicated in DNA damage response and in genome stability. It combines with other tumor suppressors, DNA damage sensors, and signal transducers to form a large multi-subunit protein complex known as BASC for BRCA1-associated genome surveillance complex. BRCA1 functions in the nuclear processes of transcription, chromatin remodeling and silencing, and in various DNA repair mechanisms. It also has ubiquitin ligase activity. It regulates key effectors that control the G2/M checkpoint and is therefore involved in regulating the onset of mitosis. It is a protein of unknown structure and has a few identifiable features. The C-terminus of BRCA1 contains two 95-residue BRCT domains. Pathological role of the protein has been identified in breast and ovarian cancer. Many alternatively spliced transcript variants have been described for this gene with only some have had their full-length natures identified. |
