NXP-2 is a unique mammalian nuclear protein expressed in specific tissues and has specific RNA binding function. NXP-2 is a member of the MORC protein family characterized by conserved structures consisting of a GHL (Gyrase B, Hsp90 and MutL)-ATPase domain at the amino-terminus, a zinc finger type CW domain containing conserved four cysteines and two tryptophans, a nuclear localization signal (NLS) and coiled-coil domains at the carboxy-terminus. The protein is localized in the nucleus and distributed to the nuclear matrix. Apart from RNA binding activity, the protein also plays important roles in diverse nuclear functions, including RNA metabolism and maintenance of nuclear architecture. A recent study has confirmed the role of the protein in SUMO-mediated repression of transcription. NXP-2 also regulates the activity of tumor suppressor protein, p53 and its localization into promyelocytic leukemia (PML)-nuclear bodies. Human NXP-2 gene is located at chromosome 21q22.13. |
