JAK2 is a member of the Jak kinase family which is ubiquitously expressed. It associates with cytokine receptors and is essential for signal transduction by mediating tyrosine phosphorylation. The protein is composed of four conserved domains, the NH2-terminal FERM domain which is required for interaction with cytokine receptors, the adjacent SH2-like fold with no known function, and a kinase-like COOH-terminal JH2 domain that is devoid of enzymatic activity but that inhibits the activity of the COOH-terminal JH1 tyrosine kinase domain. Human JAK2 gene is localized in the chromosomal region 9p24. The protein is highly expressed in spleen, peripheral blood leukocytes, testis, heart and skeletal muscle. Recent studies have implicated de-regulation of JAK2 kinase activity by chromosomal translocations in hematopoietic tumors. |
