Background
Principal Names: Calcitonin/calcitonin-related polypeptide, alpha Official Gene Symbol: CALCA Gene ID: 796 (Human) Gene Map Locus: 11p15.2-p15.1 (Human). Calcitonins (CTs) are a group of polypeptide hormones containing 32 amino acid residues which share an amino terminal ring structure, with cysteines at 1 and 7 linked by a disulfide bridge, as well as a carboxy terminal prolinamide. It is usually synthesized in the parafollicular or C cells in the thyroid gland, and also in a wide variety of other tissues, including the lung and intestinal tract. It plays an active role in calcium homeostasis. A large number of diseases are associated with abnormally increased or decreased levels of calcitonin. Alternative RNA processing of human calcitonin/ calcitonin gene-related peptide (CGRP) gene generates two mRNAs encoding the precursors of calcitonin and CGRP. It is used in the treatment of bone loss disorders because it inhibits osteoclast function. |

Application Notes
We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining. Specificity: This antibody recognizes Calcitonin, which is a 32 amino acid peptide, which can be demonstrated in C cells of the normal and hyperplastic thyroid. Staining for calcitonin may be used for the identification of a spectrum of C cell proliferative abnormalities ranging from C cell hyperplasia to invasive tumors. Staining for calcitonin in medullary carcinoma of the thyroid produces a fine granular pattern in the cytoplasm. Amyloid deposits within the tumor may also exhibit varying degrees of calcitonin activity. This antibody crossreacts with Human, Monkey, Mouse, Rat, Dog, Horse, Sheep and Pig.
