Background
Researchers have indicated that they want access to novel target antibodies quickly to aid in their research. IMGENEX is pleased to be able to offer researchers access to cutting-edge products that are still in the process of characterization/validation. IMGENEX’s ‘IMX’ antibody line is shown by ELISA to bind to the peptide used as immunogen. Investigators should empirically determine the suitability of the antibody, including optimal dilutions, for other applications of interest.
- We cannot guarantee that the yet-to-be-fully validated antibody will work in any application other than in an ELISA against the peptide used as immunogen and therefore cannot offer a refund if the antibody does not work in your application.
- The antibody is offered at a lower price compared to a more highly validated antibody.
- We are not able to provide the immunogen for the antibodies, but do provide the amino acid sequence.
These antibodies are not fully validated and we appreciate your feedback. Customer feedback is used to help generate testing methodologies which can lead to further product validation or withdrawal.
Tet1 is a member of the Ten-eleven family of proteins that convert 5-methyl cytosine to 5- hydroxymethylcytosine (1-3). Tet1 protein is an enzyme which has a molecular weight of 235kDa and has been found to be expressed in murine embryonic stem cells. It appears that Tet1 is a required protein for ES maintenance as an important epigenetic DNA modification protein. Through preferential binding to CpG rich sequences in transcriptionally active as well as Polycomb-repressed genes, Tet1 functions as a DNA methylation regulator and modulator of pluripotency markers (4). Tet1 antibody should be useful as an expression marker for differentiation and self-renewal status of stem cells. |
Antigen
Amino acids 1140-1158 [TKQKNPTQKKTKSTPSRDR] of human TET1 were used as the immunogen for this antibody.
|
Application Notes
The amino acid sequence used as immunogen is 100% homologous in human, white-cheeked gibbon, chimpanzee, orangutan and pig, 95% in new world monkey, rhesus monkey and rabbit 84% in panda, cow, dog and european pole-cat, 79% in naked mole rat and african bush elephant and 74% homolgous in guinea pig. |
