Description |
Interleukin-21 (IL-21), expressed by activated CD4(+) T cells, is a member of the common gamma-chain family of cytokines which includes IL-2, IL-4, IL-7, IL-9, IL-13, and IL-15. Its private receptor, IL-21R, is expressed on T, B and NK cells. Within the B cell lineage, IL-21 regulates IgG1 production and cooperates with IL-4 for the production of multiple Ab classes in vivo. IL-21R has been shown to activate the Janus kinase/signal transducers and activators of transcription signaling pathway upon ligand binding. Initial studies have demonstrated that IL-21 has pleiotropic effects on the proliferation, differentiation, and effector functions of B, T, natural killer, and dendritic cells. Recombinant human IL-21 produced in E. Coli is a single, non-glycosilated polypeptide chain containing 133 amino acids and having a total molecular mass of 15,463 Dalton.

Biological Activity |
Recombinant IL-21 is fully biologically active when compared to standard. The ED50 as determined by the dose-dependent stimulation of human peripheral blood mononuclear cells is < 5ng/ml. |
Endotoxin |
Endotoxin level as measured by LAL is <0.01ng/ug or <0.1EU/ug. |
Protein Content |
Protein content determined by: UV spectroscopy at 280 nm. Analysis by RP-HPLC calibrated against a known standard. Quantitation on SDS-PAGE against a known standard. |

1 ug run under (-) non-reducing conditions in a 4-20% Tris-Glycine gel, stained with Coomassie Blue. Human IL-21 has a predicted MW of 15 kDa. |
Serial dilutions of Human IL-21 (starting at 50 ng/mL) were added to Mino cells. After 66 hours, cell proliferation was measured and the linear portion of the curve was us used to calculate the ED50. The ED50 of Human IL-21 is between 0.23-0.34 ng/mL. This value is comparable to the competitor sample of 0.1-0.6 ng/mL. |
