Description |
|
CD137L is a member of the TNF superfamily and provides costimulatory signals
to T cells. It is expressed on dendritic cells, activated macrophages, and B
cells. CD137L binds to its receptor CD137 on activated T-cells, this interaction
is important for adaptive T-cell response. The engagement of CD137 by CD137L
provides costimulation to both CD4+ and CD8+ cells. Agonist anti-CD137
monoclonal antibodies also have been used to stimulate CD137 to regulate T-cell
number and functions.
The Fc-CD137L fusion protein was generated by
cloning the extracellular domain of mouse CD137L with the Fc region of mouse
immunoglobulin (hinge region, CH2, CH3 domains). Fc-CD137L fusion protein has
comparable biological activities as 2A, an anti-CD137 agonist antibody. |

Balb/c mouse splenocytes were incubated for 48-hrs on anti-CD3 (145-11C clone) monoclonal antibody coated plates in the presence of 2 mg/ml anti-CD3, 2A or Fc-mCD137L. The amount of IL-2 production induced by Fc-mCD137L was comparable to the induction by 2A antibody (Zhang N, et al, 2007). |
