DEFA6, also known as Defensin-6, is an 11kDa protein and member of the alpha-defensin grouping. Alpha-defensins are 29-42 amino acid peptides with documented innate anti-microbial activity (1).In humans, six alpha-defensins have been identified –four called Human Neutrophil Peptides (HNP) 1, 2, 3 and 4, are part of neutrophil defense mechanisms in systemic innate immunity. Human Defensin (HD) 5 and 6 are expressed in intestinal Paneth cells (2), and contribute to normal gut host defense and are dramatically increased in chronically inflamed colon (3). Paneth cells especially express DEFA6 in their lysozyme granules. Because of alpha-defensin expression in chronically inflamed gut tissue, it has also been determined and proposed that DEFA6 has potential as a marker for colon cancer (4). By RT-PCR analysis it has been determined that the highest expression of DEFA6 is found in adenoma tissue vs normal colon tissue –over 100 fold increase whereas less fold increase and frequency in carcinoma tissue overall vs normal tissue. DEFA6 greatly elevated expression patterns in adenoma vs carcinoma tissue is proposed to have more value as an indicator to discriminate between pre-malignant or earlier colorectal cancer or staging of colon cancer vs colon cancer overall (4). |

Application Notes
The amino acid sequence used as immunogen is 100% homologous in human, and chimpanzee, 83% in rhesus monkey, and white-cheeked gibbon, 78% in orangutan, and 61% homolgous in new world monkey. |
