Background GNLY (Granulysin) is characterized as a pro- inflammatory, cytolytic, 9kDa or 15kDa peptide found in granular endosomal compartments of NK cells and cytotoxic T cells (1). GNLY's cytolytic activity, in part, has been attributed to induction of cathepsin B release from target cell lysosomes which leads a damaging barrage of mitochondrial targets and a subsequent rapid cell lysis or necroptosis (2). Pro-inflammatory activity of GNLY is exhibited by induced and increased expression of inflammatory and chemoattractant cytokines such as RANTES, MCP-1 IL-1 and IL-6 on Tcells, monocytes and other cells (1, 3). Induction of CD80 and CD86 on DCs and their production of IL-6 and TNFa also illustrate its activity as a functional enhancer of the DC-Tcell synapse driving and polarizing specific Tcell responses (3). More recently, the finding that GNLY stimulation of Dendritic Cells is integrated with TLR4 and MyD88 illustrates a newly described role as an alarmin and part of the Innate Immune Response in addition to original cytolytic and adjuvant inflammatory activity (4). Antibodies to detect GNLY are valuable for assessment of its role in the varied areas of innate immunity, inflammation, chemo-attraction and cytolytic activity. |
Antigen
A portion of amino acids 70-120 from human GNLY was used as the immunogen for this antibody. |
Application Notes
The amino acid sequence used as immunogen for this GNLY antibody is 100% homologous in human, and chimpanzee, and 81% homologous in orangutan. |
